The lens has been extensively upgraded, with the new optical design featuring a significantly improved 4-stop optical image stabilization that reduces blur caused by camera shake (the old model was about 1.5 stops effective). Three image stabilization modes are available to suit different shooting conditions: standard, panning, and exposure-only. The image stabilization locks automatically when the lens is mounted on a tripod.

The shortest focusing distance is 98cm, in the old lens it was about twice that. The aperture plates are rounded, in the old version they were straight. A new type of surface material with better wear resistance.

Canon's ring-type ultrasonic motor ensures fast and near-silent autofocus.

The lens uses a traditional twist-action zoom control instead of the previous pump-action design. The new mechanism features a variable-force zoom control ring that can be used to quickly zoom or lock the zoom to the desired focal length, ensuring that the composition is maintained even if the zoom is accidentally touched.

The lens's 21-element optical construction has been completely redesigned, incorporating fluorite and Super UD elements, as well as, for the first time on an EF lens, Canon's new ASC coating, which consists of tiny air-filled nanoparticles that form a layer that reduces ghosting and flare.

The EF 100–400mm f/4.5–5.6L IS II USM features a built-in tripod ring (the tripod mount can also be removed if needed). The tripod collar locks in 90-degree increments. The new ET-83D lens hood has a slot through which polarizing filters can be rotated without having to remove the lens hood.

The lens is dust and moisture resistant, and the front and rear elements have a fluorite coating to prevent dirt from adhering.
